摘要 Abstract
❝ Cell transplantation has shown promise as a therapy for various diseases and injuries. Traditionally, cells are harvested as single-cell suspensions and transplanted into the affected area. However, this method faces challenges such as significant cell loss due to mechanical leakage, cell detachment-associated anoikis, and adverse microenvironments in the target tissues, which limit their therapeutic efficacy. To overcome these limitations, we propose the assembly of cells into three-dimensional (3D) spheroids to enhance cell retention, survival, and regenerative capacity. Our results of in vitro and in vivo studies reveal that mesenchymal stem cells (MSCs) transplanted as 3D spheroids, incorporating both cellular components and the extracellular matrix (ECM), yield superior therapeutic outcomes compared to traditional single-cell suspensions. Furthermore, we have developed a novel approach using decellularized ECM (dECM) derived from MSC spheroids, enriched with matrisome proteins and secretome. This bioactive biomaterial has proven effective in regenerative applications, as demonstrated in a mouse model of traumatic brain injury. The implantation of this 3D dECM significantly mitigated tissue damage and promoted brain repair, evident from the reduced lesion volume, alleviated neuroinflammation, decreased glial scar formation, and enhanced recruitment of neuroblasts to the injury site. In conclusion, our research highlights the potential of MSC spheroids and their derived bioactive 3D dECM as potent therapeutic agents for regenerative medicine applications.
